Three week summer training at Indian Institute of Science from 10th May 2003 to 4th June 2003.
Work profile at IIT:
Project title: Screen for a novel Agrobacterium strain that produces curdlan.
Agrobacterium, under nutrient deficient conditions, produces curdlan, a linear beta 13 glucan, extracellularly. Curdlan attracts attention because of its unique thermo gelling and rheological properties. It has got wide applications in food and drug industries.
Goal: The aim of the project is to screen for a novel Agrobacterium strain that produces curdlan of high yield and optimize the bioprocess parameters.
Responsibilities undertook in this project:
Screening was carried out to identify a new strain with good yield for curdlan. From the strains isolated bioprocess was carried out in shake flask experiments. Optimization of down streaming was done. Structural characterization of the polymer by FTIR, NMR spectroscopy, Polarimetry, Differential Scanning Calorimetry (DSC) was done.
Industrial training: A six month industrial training from 03-01-2006 to 03-07-2006 was undertaken in Xpression Biotek Pvt Ltd. The recruiters for the training are BCIL(Biotech Consortium India ltd.) and DBT(Department of Biotechnology).
The training was in the area of Clinical diagnostic kit production. I was involved in the kits like Creatinine, Urea, HDL, Cholesterol, Calcium and Phosphorous. Standardization of the kits was done. The successful kits were confirmed by quality control and production was done. Quality control was done on the manufactured kits and marketed by distributors.
Project undertook during post graduation:
Six months project undertaken at Alagappa University on the topic “Analysis of microbial contamination and stress induced genome rearrangement in Bioprocess”.
In the project, a focus was made on the detection of presence of diverse microbial contamination and the presence of genome rearrangement under starvation stress and hydrogen peroxide stress in the Bioprocess.
The project work had two parts:
1. Determination of microbial contamination
2. Determination of stress induced genome rearrangement in bioprocess
1. Determination of microbial contamination:
In the first part, investigation started with the addition of various contaminants like Bacillus pumilis (from goat skin), fungus (Aspergillus niger), incomplete sterilization, open air contamination, and improper handling of the bioprocess with the starter culture Bacillus pumilis (marine isolate). Genomic DNA of these contaminated cultures were isolated and RAPD-PCR was done.
2. Determination of stress induced genome rearrangement:
In the second part, investigation started with the induction of starvation stress and hydrogen peroxide stress to Bacillus pumilis and Bacillus firmus and the genome rearrangement is detected using RAPD-PCR technique. RAPD-PCR technique uses a primer that is not specific for a particular sequence, and also gives rapid and valid results when compared to the classical methods like plating and conventional biochemical assays in detecting the presence of contamination as well as in detecting the genome rearrangement due to stress.
